Optimalisasi Real Time PCR untuk Diagnosis Filariasis Bancrofti pada Sediaan Hapus Darah Tebal

Abstract

Penelitian ini bertujuan untuk menggunakan sediaan hapus darah tebal sebagaisampel untuk amplifikasi DNA dalam mendeteksi DNA Wuchereria bancroftidengan metode Real Time PCR. Uji Diagnostik, dengan pemeriksaan mikroskopik sebagai gold standard. Sampel adalah 63 sediaan hapus darah tebal dengan pewarnaan giemsa yangsudah diperiksa dengan mikroskop. Sampel positif terinfeksi filariasis bancrofti25 sampel, dan negatif terinfeksi 38 sampel dikumpulkan dari daerah endemikNusa Tenggara Timur. Sediaan hapus darah tebal dikerok dengan skalpel sterildan hasil kerokan sampel dimasukkan ke dalam tabung steril yang berisiphosphate buffered saline (PBS). DNA diamplifikasi dengan target Ssp I repeatuntuk W. bancrofti. Hasil dari PCR akan dibandingkan dengan mikroskopikdan tes konfirmasi yaitu tes ICT (immune chromatographic card-type). Metode Real Time PCR pada sediaan hapus darah tebal mempunyai sensitivitasdan negative predictive value yang tinggi terhadap mikroskopik. Dan hasilkonfirmasi dengan metode mikroskopik dan ICT, PCR pada sediaan hapusdarah tebal memberikan hasil sensitivitas, spesifisitas, positive and negativepredictive value yang tinggi. Korelasi Spearman menunjukkan korelasi yangkuat antara mikroskopik dan PCR pada sediaan hapus darah tebal (r = 0,937).Dan korelasi negatif antara nilai Ct dengan densitas mikrofilaria (r = -0,726).
